to compare CsBz flies and native flies comparing the sense of smell between the two hypotheses is the smelling capacity in the native fruit fly is stronger than CsBz and this is because of evolution Do we understand what is evolution, mutation and natural selection?

preparing a device to quantify the sense of smell using 1% Agar solution adding 20 ml in each plate allowing it to cool down and solidify using distilled water on both sides to calibrate the device we expect the equal distribution of fruit fly larvae on either side because distilled water doesn’t have any order using Isoamyl acetate that is the bananas smell as odour expectation Larvae we will move towards isoamyl acetate instead of distilled water. what is written on the board more number of flies going towards water? is the water acting as an attractant? or Isoamyl Acetate acting as a repellent? distilled water is used as a control in this experiment!

in the lockdown, due to Covid situation, we don’t have access to Labs To Agar and Isoamyl acetate can we still do the same experiment using rava in place of agar to provide a smooth surface to the Larva using banana juice in place of Isoamyl acetate why are we using the second instar larvae how to identify the second instar larvae?

Discussed about comparison between Native fruit flies and CsBz fruit flies. One of our hypothesis is

_Hypothesis :- Native fruit flies smelling capacity is much more better than CsBz fruit flies. _

Because CsBz fruit flies already have food (Standard corn meals agar media). They don’t want to find any food. But in the case of native fruit flies have to find food their own self. - This based on make a hypothesis and some of them are mutated in CsBz flies.

Example of Experiment Test
Olfactory test - Smelling senses of fruit flies

In Lab taken for this experiment Agar plate, IAA (Banana essence smell), Paraphinoil (With out any smell), 2nd inster larvae of Drosophila melanogaster.

While started experiment check olfactometer works or not using distil water. Both side put Distil water in a Agar Petri dish. Middle of petri plate put 10 larvae. After 2minutes later observed larvae are move random - olfactometer is working. Other ways only larvae moving one side (Example A) - olfactometer have some problems. -

*IAA / Distil water *

_Expectations :- _
•7 Larva move towards IAA.
•3 Larva move towards Distil Water.

Repeated Experiment

_Expectations :- _
•6 Larvae move towards IAA.
•4Larvae move towards Distil Water.

*Home Experiment *
Olfactory test

_Objective :- To Finding Olfactory Sensitivity of 2nd Instar Larva of SLBK A (one of single line culture) _

Agar - Replaced Rava
Petri dishes - 12 cm plate

IAA - Banana Paste
5 Different Concentration -
•1 Table spoon BP + 1 Table spoon water
•1 Table spoon BP + 2 Table spoon water
•1 Table spoon BP + 3 Table spoon water
•1 Table spoon BP + 4 Table spoon water
•1 Table spoon Bp + 5 Table spoon water

Paraphin oil - Tap water.

_Experiment _ : To make a Boil Rava Paste 100mL for each 3 normal plate 12cm. Each 3 plate 30mL. After 1and 1/2 hour for kept it cooling to cover with other plate for not enter other files.

Take one Rava plate and to check our olfactometer work or not. Put both opposite side one drop tap water and middle of Rava plate add 10 - 2nd instar larvae. After 2 minutes later observe larvae are equal movie each side like A side 5 & B side 5 larvae. Our olfactometer works. If larvae are moving only one side our olfactometer have problems. - Control Test
