Discussion had started what’s the standard way to prepare TRSV Culture Medium as Sachin has also tried to make 50ml TRSV medium but that Medium has not solidified after 1 and half days…

We had tried to find out where could be the error ? Sachin told us the components that he used the method that he followed…

After that Aswathy and Isha had also shared the method that they are following in order to make medium.
This is the collage of all the components,this picture was shared by Isha…

We had also seen a vido which is made by Isha
We got an update from CUBE home lab worli Mumbai, where Isha is maintaining two single line Cultures of Fruit flies and the primary objective is to identify single line Cultures…
Above is the picture taken by Isha and lower one is the picture of wing taken by Aswathy from Bottukavadu kerala single line culture…

We can clearly see that costal vien is absent in the wing, is it because of handling error or is this wing doesn’t have costal vein?.

Isha showed us three photos of wing from the same single line Culture fruit flies but costal vein was not seen in any of the wing.
We had discussed how to take good photos of wing or can we try to take picture of wing without removing it from the body of fruit fly.

Isha immediately went and took one picture of wing intact with fruit fly and costal vien was very much visible so actually it was the handling error due to which costal vien was not visible…

Above picture is the wing of CsBz Drosophila melanogaster and lower one clicked by Isha .
We had also discussed the optics, how to take good picture of a wing and a Head? What could be the arrangement of touch light, paper , transparent sheet amd phone camer?
From which sided the light should be flashed?

The picture would be clear if the light source is from below in case of wing and whereas in case of head light source either from the top or from side.

Some terms were discussed like Dorsal, Ventral, Anterior, posterior, Proximal and Distal

Which is the dorsal side of fruit fly and which one is the dorsal side of a leaf?.
On what basis we are saying that the back side of animal is Dorsal?.
How do we say what and how are me measuring the growth of the Nail?..

Can we say this in terms of Proximal and Distal end?.
Proximal which is in proximity or close to the body, distal is the end which is far from the body.

How do we say that in the context of Nail growth?.
After the discussion we have made the conclusion that
Nail growth is measured in between the proximal end of nail and proximal end of mark
