Biodiversity at your Doorstep

Pavement of road has manhole lid covers, which have two depressions on it, during rainy season it will be filled with water.

Will that small well have some organisms? Can we make it a model to study biodiversity of micro-organisms at our doorstep?
What kind of micro-organisms can be present in it?
Please make some propositions and post it in reply section below, by clicking “Reply” icon.

Micro-organism present in soil sample:

Collection of Water sample:

We need to collect the water sample present in the well and observe under the microscope to find out what organisms are present in it?

What will be design of an experiment to collect sample?
What will be design of an experiment to observe the samples?

DAY 1 Experiment 1 :-
AIM - To study Evolution of organisms
Firstly we went around the school campus looking around for soil samples for our experiment.
After some digging we found 3 samples
1st - it was found near the canteen area at around 9:45
2nd - it was found just adjacent to CP block at 9:55 precisely
3rd - this one was covered with algae and found near the Hostel area at 10:03
then we took 4 bottles and filled them with dechlorinated water
after that we took 1/3 of all the soil samples and mixed them with 1 bottle of dechlorinated water each
then we used a dropper to pour 2-3 drops of milk in 3 of the bottles
for the micoorganisms to be enlarged by dividing and enrichemnt we had to leave the solution for 24 hours
We already had 1 sample which was already enriched . this sample was taken from Mumbai around several weeks before
we will observe that sample tommorow to comapre the diversity of organisms
The challenge was how do we measure the size of our microbes. For this we took our plastic scales under a microscope to find out measurement of a micron which is 1/1000 milimeter = 0.001 milimeter
atlast we observed round worms and rotifers under a microscope

Day1 Experiment 1

Aim:-

To study evolution of organisms

Procedure & Observation:-

We went around the school campus looking around for soil samples. The soil had to be collected from a shadowed place and the soil should be wet. Three types of samples were found:-

  1. We found it near the canteen at around 9:45 am.
  2. We found it adjacent to the CP Block exactly at 9:55 am
  3. We found it near the hostel and was covered in algae. It was collected approximately at 10:00 am

Four bottles were taken and were filled with dechlorinated water. A third of every soil sample was taken and was mixed with the bottles of dechlorinated water. 2-3 drops of milk were poured into the bottles with a dropper for the enrichment of the microbes. This apparatus was to be left for 24 hours and will be observed tomorrow. We had a sample which was already enriched, ie, the microbes had already been cultured and had grown. This sample was taken from Mumbai before several weeks.

As a side activity, we measured our scales under a microscope to find out the measurement of a micron.

We also observed roundworms or nematodeunder a microscope. Roundworms are found in the intestine and the soil

Day 2 Challenge 2

Aim:-

To modify a web cam into a digital microscope

Procedure and result:-

Today we had a challenge of making a microscope out of a web cam. Though at first we all were really confused on how to approach it, we eventually found out a way. We started the progress by first making a stand for the microscope. First we made a three layered base on which we attached a vertical board with a hole in the centre. On this board we attached another piece of cardboard cut in a shape to support the microscope. To hold the microscope we decided upon using two equally cut pieces of cardboard with two holes to fix the microscope.

After the stand was completely build we had to start with converting the webcam into a microscope-our main challenge. To build the microscope we inverted the optics but a problem we faced was that the inverted optics were not focusing very well. This took us a lot of time to finally make the optics focus to complete the microscope . But now this microscope is now completely functional.

While watching the leaf sample from cp block bushes I found round worms in it. The movement of it was amazing to watch so I have also shared it with you.

A small clip of a sample we were provided by the STEM Games. This sample is from Mumbai and was cultivated for about 20 days.https://stemgames.metastudio.org/uploads/default/original/2X/7/79a7b28abf86e30fcc748215ed80578e6ba357db.mp4
