Date of Culture : 16th April 2021

Objective; is to find D.melanogaster (One of the species) around my native area.

•4 single line culture A, B, C, E.
•newly made sub cultures - A15, B15, C15, E10 made on 16th April 2021
•~30 - ~35 fruit flies in each 4 bottles.

Larvae Gut Color Change Behaviour
Observed in Single Line Cultures Bottles

When added Apple vinegar (Color vinegar) while media preparation instead of adding white vinegar. With in few days (3-4 days) media color completely change in to dark brown. This media feed all 3mm sized larvae gut color changes to brown except SLBK C culture…

Why Only Few Larvae Gut Showing This Kind Of Color Change?

This behaviour only can see in 3rd instar Larvae , but not all larvae.

The acidic region of the larval Drosophila midgut. (A) Midgut of an intact Canton S larva fed mCresol purple pH dye (red pH < 2.4 yellow pH 2.5–8 purple pH > 8); (B) Excised midgut of a Canton S larvae maintained on m-Cresol purple pH dye showing five regions of pH. The anterior acidic region is spatially distant from the posterior alkaline region after midgut dissection (B), but lies parallel in the intact larva (A).

The H + V-ATPase complex is required for acidic pH generation. Midgut pH was assessed using Thymol blue pH dye (red pH < 2.4, yellow pH 2.5–8, blue pH > 8). Canton S and parental controls (A–C,E,G) all maintain a region of pH 2 (orange/red staining) in the acidic region (annotated by white arrows), whereas acidity is reduced in the vha100-2 (D) and vha100-4 (F) knockdown lines (yellow staining), but not the vha1005 knockdown line (H).
