Is it possible to perform Olfactory Assay at home to compare the olfactory sensitivity
Discussion went on that how would be go about performing olfactory assay?.
In between the discussion got diverted as Isha From worli Mumbai joined the cube chatshaala and we got to know that a new CUBE HOME LAB is been made.
Isha has started maintaining single line Cultures of Native fruit flies…

One of the Cubists from Delhi is also trying to trap fruit flies by keeping an open bait in balcony on 2nd Floor…
It’s been two days and not a single fruit fly has visited the bait.
So discussion went on deeper and enabled us to understand what’ would be the further plan and there were suggestions which would definitely help Raj in order to get fruit flies.
Bottles on the whiteboard shows that the discussion was roaming around how to isolate single gravid female fruit fly from a population.
How to differentiate Male and female Fruit fly?.

What would be the next step after isolating one Gravid female fruit fly .
The TRSV medium bottles will be prepared in order to culture fruit fly and to Culture the progeny of the fruit fly.
The discussion again got back to the original question that’s to perform olfactory Assay at Home…
What’s the need of performing Olfactory assay?.
What all questions we can address?.
One of the long term objective is to compare the olfaction sensitivity of Standard CsBz strain of Drosophila melanogaster and Native Drosophila melanogaster
But why?. would there be any difference in their olfaction sensitivity?.
How do Native fruit flies survive in an open environment where they have to hunt for feeding, mating and egg laying. Whereas on the other hand CsBz fruit flies are getting everything ready and it’s been happening since 70 years…
Can we make out something now?.There will be mixed variety (Best ,better, mediator, good, less ,worst olfaction) and only those species would survive who has the best or better or mediator.
On the other hand every Species will survive nonetheless as the food is already available…
But the CsBz was the Native at first?.how do they get Changed or Mutated?.
What’s Mutation?. Can we make out the difference among Mutation, Variation, Evolution and Natural selection?.
How do we understand the concept of Mutation at Molecular Level?.
Can Mutation be happened during the replication or Translation?.
So the discussion went on deeper and we had proposed a model what could happen at Molecular Level?.
Blue fruit fly is having better sense of smell?.and how does change in Nucleotide sequence would lead to mutations and hence the progeny becomes less sensitive to smell?.
If there’s a chance that fruit flies can have different sensitivity so how do we quantify it?.
Olfactometer can be used in order to compare the olfaction in between CsBz and Native
Drosophila melanogaster.
Since Aswathy has been maintaining four single line cultures at Home and Aswathy has proposed that the can compare the olfaction in between two single line cultures by keeping the fact that they could be of two different species…
We had also discussed about the Lab method and than substitute the materials in order to perform olfactory assay at home.
Base of plastic bottles can be use in place of petri-plate
Rava can be use in place of an Agar
Banana Paste can be an alternative to IAA.
Well water to be replaced with Distilled water.
