Picture presentation: How well we are able to observe things?.

Same Another observation made by Aswathy from one of the bait Culture of fruit flies is that Red color is seen at the posterior end of Larvae…

Discussion revolved around why this red color is seen in these Larvae?.

Some presumption made by the Cubists which are as follows

  1. Red color is seen because larvae are feeding on tomato and tomato is red in color, hence red color is seen in the mid gut.

  2. These could be of different species

  3. Comparison was made with Moina as moinas turn red when undergone low oxygen level condition, is color change can also be seen in larva of fruit flies?.

  4. may be a varient

  5. Another presumption made that due to to ph of tomato.

How do we varify all these presumption?..

https://www.researchgate.net/publication/303774643_Molecular_mechanism_and_functional_significance_of_acid_generation_in_the_Drosophila_midgut. Midgut pH assays. pH was measured in the larval midgut using ymol blue, m-Cresol purple, Cresol red, Bromocreosol purple, Bromothymol blue, Phenol Red and Congo red indicator dyes (all purchased from Sigma- Aldrich). Indicators were added to melted Drosophila diet (0.1% w/v), immediately mixed, and allowed to cool to room temperature. Larvae of the appropriate genotype were added, and aer 2 hours the midgut excised in Schneider’s insect medium (Invitrogen). Micrographs were taken immediately using a Sony CyberShot NEX-C3 mounted on a Leica stereo microscope, as pH remains stable for only a few minutes aer dissection. Images were processed using Adobe Photoshop CS5.1. Where noted, the diet was also supplemented with 1 mM Omeprazole (Sigma-Alrdich) or 100 μ M acetazolamide (Sigma-

Bait(where one slice of tomato) was kept on 17th January 2021 in order to trap fruit flies from Native(Moradabad UP).

One Gravid fruit fly got trapped on 25th January 2021(on 9th Day).

First Larva(4mm) was seen on 03rd February 2021

First pupa was seen on 06th February 2021.
Objective: to study life cycle of fruit fly

Approx 5mm size Larva seen on the top of bottle today (06th February 2021)
