NEMATODES
CUBE Home Lab Kolkata - Batul shared an experience How to isolate nematodes from soil using potato slices adding one drops of milk. 15 days we can able to maintain this culture in that potato slice!. Using safety pin for to transfer nematodes from one slice to another new slice.
Can we make single line culture of Nematodes Is it easy or difficult!?
According to Batul we can’t much able to see this worms
in naked eyes. We need to make sure only one worm we are transferring in to new potato slice, then only we can say it is single line culture of nematodes. In that case we can take some sample from mixed culture of a worms and looking under the microscope we can identified worms try to take one nematode with the help of safety pin. Transfer in to new potato slice.
Batul made a single line culture of nematodes on last year December 2020.
MICROWORMS
Observed First Microworms Present Inside the cultures on 24/4/21 made culture SLBK A bottles.
No larvae No Flies No pupae (10 days later after transferring ) No eggs
Worms noticed when looked closely And collaborator @Drishrant tried comparing it with Nematodes
QUESTION - IS A MICRO WORM AFFECTING THE GROWTH OF FRUIT FLIES?
HYPOTHESIS - MICRO WORM IS NOT CAUSING THE DEATH OF FRUIT FLIES.
Criteria of good culture ?
Transfer date - 19/7/21
7th Day
day 1 - 30 flies (8/30 gravid )
day 1 - 1 gravid flies
day 2 - 1 fly lays 50 eggs (in 1 day )
7 day (each day 50 eggs ) - 7 X 50= 350 we have 8 flies - 8 X 350 +2800
SLBK A21 SLBKB20 SLBKC21 SLBKE16 Transfer date -19/7/21 no of flies transferred 30 no of flies 22 no of pupae 15 no of larvae 20 Microworms present inside of Media
MICROWORMS
“Micro worms lifecycle - 3 days day 1 - Egg of Micro worm day 3 - Micro worms day 4 - new micro worm will lay an egg 60 eggs in a single day supposedly 10 eggs were transferred 21/7 - 10 microworms 1 micro worm = 60 eggs 10 micro worm = 600 eggs 7th day - 7x 600 = 4200”

Microworms
How will you get eggs everyday? Those 60 eggs will become adult in 3 days and then may lay eggs
These guys are incredibly easy to culture. Make some plain instant oats (I like a pretty thin consistency), let it cool completely and add the oats to a plastic container. Remember some air holes! Add your culture and give them some time to populate – you will be scraping them off the sides in no time.Live Culture: Microworms | Simply Betta
600 microworms on 4th day which means 60 eggs by 600microworms and we should multiply 60 with 600
60 eggs by one microworm and 600 microworms will give 60* 600 eggs
“We can use this formula = ( n-5) *( 60 * number of microworms transferred )for calcultating number of microworms for 10 microworms placed intially in bottle but when we think about the eggs layed by 1st generation 600 microworms i am not able to think for a formula for that.”

How make a design of Experiment to test when is the worm started to presenting culture bottles!?
