Summary of chatshaala discussion.

Discussed model systems : Fruit Flies, Nail regeneration.

Yesterday we were discussed about activity pattern of Fruit flies inside a culture bottles. Do fruit flies sleep at night!!?

Experiment : Checked flies culture 4 bottles, morning 5 times and night 5 times want to provide/disproved flies do flies sleep at night!

Design of the Experiment : 4 Fruit flies freshly made culture bottles taken for this experiment (that will help us to understand activities of fruit flies clearly.Only 6 days old culture bottles, most of the flies newly emerged flies (15/04/2021). Kept this bottles inside home (Hall). There was light normal conditions (not much darkness/lightning). When was taking videos and photos, only that time was on the light in there.

Experiment : Taken 5 set videos of that culture bottles at morning & night. Totally ~100 flies (A= ~30, B= ~20, C= ~25, E= ~20)

Observation : Day 1st

Morning :-
7:00am - 80% not active
7:30am - 50% active (flying + walking)/ 50% not active
8:00am - 50% active (feeding + walking + flying)
8:30am - 80% active (feeding + walking + flying + courtship behavior)
9:00am - 100% active (feeding + walking + flying + courtship behavior).

Night :-
09:30pm - 50% not active + 50% active
10:00pm - 75% not active + 25% active (walking + flying + playing under cotton plug)
10:30pm - 80% not active + 20% active (walking + flying)
11:00pm - 90% not active + 10% active (walking, but not continuously)
11:30pm - 100% not active.

Planing to take videos continuously 4 days will get a pattern. That will help us to plotting graph.

Circadiam rhythm graph was showed that some of fruit flies were visited fruit bait at night 12:00am time and continuously presented that flies till 6:00am(Night active flies). What are they doing on bait!? Are they resting/sleeping on bait!? or they doing some activities on bait!!?. How can we understand that those flies active or not!?

Made a design of an experiment that we can trap those night active flies - Nocturnal flies. And culture them in a media bottles. Can start to observing flies at Day and Night, what could be their activities day and night. Nocturnal flies will sleep at night???:thinking:

Aastha & Hithesh intersected to study about this nocturnal flies.They made a design of an experiment

2 Hypothesis :
•Fruit flies is rest on bait at night.
•Fruit flies is active on bait at night.They are natural variants.

In this Experiment only we need Bottle & Fruit (Banana / Tomato). Cut fruit very small pieces and put in to bottles. Keep this bottle at kitchen garbage bin & kitchen window side. To chek at night is there any flies present inside bait kept bottles. When the flies will present at bait, then closed this bottle using cotton plugs. If you want to culture them, we can transfer those flies in to media bottles. Other ways the bait kept bottle is enough to observe the flies activities at day and night.
